Zydus Lifesciences Limited and Formycon AG have reached a major regulatory landmark with the US Food and Drug Administration (USFDA) accepting the Biologics License Application (BLA) for FYB206, a proposed biosimilar to the blockbuster oncology drug Keytruda (pembrolizumab). To the companies’ knowledge, this marks the first publicly announced BLA acceptance by the USFDA for a proposed Keytruda biosimilar.
Streamlined Development Pathway Without Phase III Efficacy Trials
A defining characteristic of the FYB206 development programme is that it proceeded without a comparative Phase III efficacy trial. Following an agreement with the USFDA in February 2025, this streamlined pathway significantly accelerated development and lowered financial investments while strictly upholding the high scientific benchmarks required for biosimilar authorization.
Under the Biosimilar User Fee Amendments (BsUFA), the USFDA review window targets a final decision within 12 months, pointing to a potential regulatory outcome in the third quarter of 2027. Additionally, the regulatory dossier has been officially submitted to Health Canada.
Strategic Partnership and Commercialization Rights
Under an exclusive licensing and supply agreement finalized in December 2025:
- Zydus Lifesciences holds exclusive commercialization rights for FYB206 across the United States and Canada.
- Formycon AG managed development, completed regulatory filings, and retains responsibility for product supply, alongside holding global rights and participating in commercial gross profits.
- The USFDA’s BLA acceptance serves as a contractual milestone, triggering a formal milestone payment from Zydus to Formycon.
Significance of Pembrolizumab and Market Potential
Keytruda (pembrolizumab) is an immune checkpoint inhibitor targeting the PD-1 receptor to assist the immune system in detecting and destroying cancer cells. As one of the world’s top-selling biologic medicines—reporting worldwide sales of USD 31.7 billion in 2025 and USD 16.40 billion in the first half of 2026—it represents a cornerstone treatment across multiple cancer types.
While the BLA acceptance is a major procedural step, FYB206 remains an investigational product pending final regulatory approval and the resolution of any relevant intellectual property considerations. If ultimately approved, the biosimilar could broaden patient access and affordability for vital cancer treatments in North America.
