
Pharmaceutical company Eli Lilly has announced a major success in its clinical development program for orforglipron, a new oral medication for obesity and type 2 diabetes. The drug met all primary and secondary endpoints in the ATTAIN-2 Phase 3 trial, paving the way for global regulatory submissions.
Orforglipron’s Mechanism and Potential
Orforglipron is a once-daily, oral GLP-1 receptor agonist. Unlike many current GLP-1 drugs that require injections, orforglipron is a small-molecule, nonpeptide medication, which allows it to be taken as a pill. It works by mimicking the hormone GLP-1, which stimulates insulin release, slows digestion, and promotes a feeling of fullness. This dual action helps reduce food intake and leads to significant weight loss and improved blood sugar controlAccording to Dr. Louis J. Key Findings from the ATTAIN-2 Trial
The ATTAIN-2 trial was a large-scale, 72-week study involving over 1,600 adults with obesity or overweight and type 2 diabetes. The trial compared various doses of orforglipron (6 mg, 12 mg, and 36 mg) against a placebo.
The results demonstrated significant benefits for patients taking orforglipron:
- Weight Loss: Patients on the highest dose lost an average of nearly 23 pounds over 72 weeks.
- A1C Reduction: The drug lowered A1C levels by 1.3% to 1.8%, helping a majority of patients (75% on the highest dose) reach the American Diabetes Association’s A1C target of 6.5% or lower.
- Cardiometabolic Improvements: The drug also showed positive effects on other cardiovascular risk factors, including non-HDL cholesterol, blood pressure, and triglycerides.
Kenneth Custer, an executive at Lilly, stated that the company is “moving with urgency toward global regulatory submissions” to meet the needs of patients. If approved, orforglipron could redefine how obesity is treated by offering a convenient, once-daily pill that can be scaled globally to reach more people.
